After reading this thread - http://forums.malwarebytes.org/index.php?showtopic=5736 I'm assuming the reason given for Malwarebytes not being able to be used from a live CD is more than likely the same reason that combo fix wont work...
persistent updates are defiantly an advantage of a bootable USB, but you can update your virus definitions temporarily on the boot CD. So there is still no difference to functionality.
I haven't bothered to make a hiern's bootable USB yet, but have bootable BT3 + BT4 USB. A hiern's Bootable USB is definitely the way forward as regards easy of use, but makes no difference to functionality.
