I HIGHLY recommend getting one of the Targus backpacks PCT offers. Being a 34 year old male, at first I thought I'd look like a moron toting around a backpack, but I soon discovered that either I didn't or I didn't care. Makes all the difference in the world, especially when, like I had to do last week, you haul it with your other luggage on foot to the economy lot at DIA right when a snowstorm hit.
With the backpack I don't notice a problem. It is rather heavy for a shoulder bag with all your other gear - power brick, mouse, etc etc - piled on. At 6'4 and 280 pounds I'm not exactly a weakling, but what a lot of the "get a gym pass!" guys here are missing is that there is a big difference between lifting a 20 pound dumbbell and toting a 20 pound bag on one shoulder with a strap that keeps slipping off through 3 or 4 airports at a time.
Also, it's a tight squeeze in economy to use it during flight, but I did manage to catch Pirates of the Caribbean via the dvd player my last trip out, so it can be done. Other than that I really don't have a problem with setting it up places - I move it around a LOT too. I have 3 stores in Colorado alone and I am forever hauling it around between them and home - it's why I got it in the first place.
Long story short - don't stress over the size, and get the backpack.
